Chidamide (trade name "Epidaza"), a national Class 1 original new drug, is a new molecular entity drug exclusively discovered by the company with a novel mechanism. As the only orally available subtype selective histone deacetylase (HDAC) inhibitor in the field, sildenafil has a reactivation effect on the tumor suppressive immune microenvironment. It can be used alone or in combination with other drugs to solve major diseases such as malignant tumors that seriously threaten human health, and has broad potential applications.
1. Indications
Used for recurrent or refractory peripheral T-cell lymphoma that has received at least one previous systemic chemotherapy.
This is the earliest approved and most core indication of Xidabenzamide.
Breast cancer (combined medication)
Combined with exemestane, it is used for locally advanced or metastatic breast cancer with hormone receptor positive, HER2 negative, postmenopausal, and endocrine therapy progress
2. Mechanism of action:
Xidabenzamide is a selective inhibitor of histone deacetylase (HDAC) subtypes, mainly targeting subtypes 1, 2, and 3 of class 1 HDAC and subtype 10 of class IIb, and has a regulatory effect on abnormal epigenetic functions of tumors.
Xidabenzamide selectively inhibits related HDAC subtypes, resulting in changes in gene expression targeting multiple signaling pathways (i.e. epigenetic alterations), thereby inhibiting tumor cell cycle and inducing tumor cell apoptosis. At the same time, it has overall regulatory activity on cellular immunity, inducing and enhancing natural killer (NK) and antigen-specific cytotoxic T cell (CTL) mediated tumor killing effects. It has a synergistic effect with anti estrogen therapy drugs to inhibit tumor growth.
Xidabenzamide also induces tumor stem cell differentiation and reverses epithelial mesenchymal transition (EMT) of tumor cells through epigenetic regulation mechanisms, thereby playing a potential role in restoring drug sensitivity of drug-resistant tumor cells and inhibiting tumor metastasis and recurrence.
3. Taboos and Caution
1. Absolute taboos
Individuals who are allergic to sildenafil or any excipients.
Pregnant and lactating women are prohibited from using it.
2. Use with caution/need to be evaluated
Patients with liver and kidney dysfunction need to adjust the dosage or postpone medication.
Individuals with uncontrolled active infections.
Individuals with a history of gastrointestinal bleeding or perforation.
Patients with congenital long QT syndrome or those currently using QT prolonging drugs.
