Sunvozertinib: Usage and Dosage

Sunvozertinib is an oral targeted therapy indicated for the treatment of specific subtypes of non-small cell lung cancer (NSCLC), such as those with EGFR exon 20 insertion mutations. Its standard dosage is 300 mg administered once daily, with the tablet(s) swallowed whole; administration may be with or without food. The specific regimen must be strictly followed as directed by a physician, and the dosage adjusted based on the patient's tolerability.

Usage and Dosage

1. Routine Dosage

The recommended dosage is 300 mg once daily (e.g., two 150 mg tablets). Administration should be at a fixed time each day to avoid missed doses.

If a dose is missed by ≤ 12 hours, take the missed dose immediately.

If more than 12 hours have passed since the missed dose, skip the missed dose and resume the regular dosing schedule the next day. Do not double the dose to make up for a missed one.

2. Dose Adjustment

Hepatic dysfunction: Dose adjustment is required for patients with moderate to severe hepatic impairment, with the specific regimen determined by the physician following a comprehensive assessment.

Adverse reaction management: In the event of grade ≥ 3 adverse reactions (e.g., diarrhea, rash, thrombocytopenia), suspend medication until symptoms resolve; a subsequent dose reduction to 250 mg once daily or 200 mg once daily may be required based on clinical evaluation.

3. Medication Precautions

Concomitant use with potent CYP3A inducers (e.g., rifampicin, phenytoin) is not recommended, as such use may reduce the therapeutic efficacy of Sunvozertinib. If co-administration is medically necessary, dosage adjustment must be performed under the guidance of a physician.

Regular monitoring of complete blood count, liver and renal function, and electrocardiogram (ECG) is required during treatment.

Hypersensitivity to any component of Sunvozertinib.

Precautions

Caution is advised in pregnant women, lactating women, and patients with pre-existing cardiac disease.

Adverse Reaction Management

Common adverse reactions include diarrhea, rash, stomatitis, and anemia. Most are mild to moderate in severity and can be alleviated with symptomatic treatment.

Immediate discontinuation of medication and medical attention are required if severe adverse reactions occur, such as interstitial lung disease, QT interval prolongation, or heart failure.
