Olverembatinib is a targeted therapy agent primarily indicated for adult patients with chronic myeloid leukemia (CML) or acute lymphoblastic leukemia (ALL) harboring specific gene mutations, especially those who are resistant or intolerant to existing tyrosine kinase inhibitor (TKI) therapies. By inhibiting the activity of abnormally activated BCR-ABL1 tyrosine kinase, this drug blocks the proliferation signals of leukemia cells.

Indications and Patient Populations

1. Chronic Myeloid Leukemia (CML)

It is indicated for adult patients with Philadelphia chromosome-positive (Ph+) CML in chronic phase or accelerated phase who carry the T315I mutation. These patients develop resistance to conventional TKI agents (e.g., imatinib, dasatinib) due to gene mutations, and olverembatinib can selectively inhibit the activity of the mutant kinase.

2. Acute Lymphoblastic Leukemia (ALL)

It is indicated for the treatment of adult patients with relapsed or refractory Ph+ ALL, particularly those with the T315I mutation who have failed other treatment regimens.

Mechanism of Action and Clinical Value

Targeted therapy

Olverembatinib exerts its effect through highly selective inhibition of BCR-ABL1 tyrosine kinase (including the T315I mutant variant), thereby blocking the key signaling pathways that drive the abnormal proliferation of leukemia cells.

Overcoming drug resistance

The T315I mutation is the primary cause of resistance to first- and second-generation TKIs in CML and ALL patients. Olverembatinib demonstrates significant inhibitory activity against this mutation, filling the gap in clinical treatment.

Precautions for Medication Use

Prescription-based administration

Olverembatinib is a prescription-only medication. The specific dosage and treatment course must be adjusted based on the patient’s genetic test results, disease stage and tolerance. Patients shall not self-administer the drug or modify the treatment regimen without medical supervision.

Common adverse reactions

Adverse reactions include thrombocytopenia, anemia, abnormal liver function, etc. Regular monitoring of blood routine, liver and kidney functions is required during treatment.

Contraindicated populations

Individuals allergic to any component of the drug, as well as pregnant or lactating women, are contraindicated from using this medication. Strict contraception should be practiced during the treatment period.
