The recommended standard dosage of Lietinib is 150 mg once daily, to be taken on an empty stomach (1 hour before or 2 hours after a meal). The specific dosage should be adjusted based on the patient’s condition, tolerance and physician’s assessment, and must not be increased or decreased without authorization.

I. Standard Dosage Instructions

Routine dosage: 150 mg orally once daily, administered on an empty stomach to ensure optimal drug absorption.

Treatment cycle: A typical treatment cycle is 21 days, which may be adjusted according to the specific therapeutic regimen (e.g., combination with other drugs or radiotherapy).

II. Dosage Adjustment Factors

Adverse effect management

In case of grade ≥3 adverse reactions (e.g., severe rash, diarrhea or abnormal liver function), the physician may consider treatment interruption or dosage reduction (e.g., adjusted to 100 mg per day).

For patients who cannot tolerate low doses, the dosage may be further reduced to 75 mg per day.

Hepatic and renal impairment

Patients with moderate to severe hepatic impairment require dosage reduction; dosage adjustment is generally not needed for patients with renal impairment, but close monitoring is essential.

Drug-drug interactions

When co-administered with potent CYP3A4 inhibitors (e.g., ketoconazole), the dosage of Lietinib should be reduced to 75 mg per day; when co-administered with potent CYP3A4 inducers (e.g., rifampicin), the dosage may need to be increased to 225 mg per day.

III. Precautions for Administration

Strictly fasting: Do not take the drug with food; avoid eating within 1 hour before and after administration.

Regular monitoring

During treatment, patients should undergo regular monitoring of liver function, blood routine and cardiac function to promptly identify adverse effects.

Missed dose management

If a dose is missed and the next scheduled dose is more than 12 hours away, the missed dose may be taken; otherwise, skip the missed dose and never take a double dose.

IV. Special Population Instructions

Elderly patients

Age alone does not require dosage adjustment, but the dosage should be tailored based on the patient’s overall physical status.

Pregnant and lactating women: Lietinib is contraindicated, as it may cause harm to the fetus or infant.

V. Medical Guidance

Lietinib is a prescription-only medication. Prior to use, it must be evaluated by an oncologist. During treatment, patients should attend regular follow-up visits and report any discomfort symptoms. Any dosage adjustment or treatment discontinuation must be performed in accordance with the physician’s instructions, and shall not be decided by the patient without authorization.
